I have a aeromotive AFPR, but I am waiting on a fitting to install it. In the meantime I think my fuel pressure is off now because of the 255 I have installed. With the gauge in hand, what vacuum lines go where so I can pressure test it?

Well the gauge shouldn't be in your hand... it should be tapped somewhere on your fuel line. Then you just unplug the vacuum line to the fuel pressure regulator and cap it with your thumb and read the number on your gauge. I'm a little confused by your question about hooking up vacuum lines

Well, I have an AFPR not installed yet, was just wondering if I could hook up one of the vac lines to it to check fuel pressure.

Fuel pressure isn't measured by any of the vacuum lines, its measured my the actual pressure inside the fuel lines. I'm assuming that your gauge is attached to your regulator? If so just wait until the get your fittings and then install the regulator/gauge and the get it set. Since you have a 2g you'll want to set your base fuel pressure at 43 psi.
