I just got my friends block back from the machine shop and am starting his build. As far as I know i need to check ptw clearance, main bearing clearance, crankshaft endplay, and piston to valve clearance. Is there anything Im missing? Any input is great!

why was the block at the machine shop?

also it couldn't hurt to check the block deck for warpage, clean out the oil squirters, clean all oil galleys, check rod bearing clearance, piston ring gap, balance shaft bearings. and im sure there is more but i can't think of it

Chase down with a tap deck and main holes to make sure threads are nice and clean. If it is a 2g block, then you shoul have spring/ball loaded oil squirters. Make sure oil squirters' passages are clean, you can take a can of wd40 with a straw attached to it, connect the straw to the inlet of the squirter and slowly press on can's nozzle to inject wd40, once enough pressure is build up the spring will collapse letting the ball pass wd40 through the squirter and will see a jet of wd40 exiting the squirter. Doing so will clean your squirters and test them at the same time, you do not want squirters to free flow oil in case where spring is damaged or a debry is stock causing ball not to be seated all the way, and at the same time you do want squirters to open under preassure.
If you have 1g block, then I am not sure if those squirters are spring/ball loaded. I did hear that 1g squirters come loose if not fastened properly.

first thing you do after getting the block on the engine stand is wash it!

do not tap the head bolt hole, this can/will thin the threads out and weaking them, use a old head bolt and make a thread chaser, take your cut off wheel and cut 3 grooves across the threads and hand tool only scew them in and out blowing out with compressed air and break clean as needed.

also make sure you clean the crank and gun brush out the oil galles in the crank
