I don't see the point. It's to take over under boost when the stock injectors can't keep up. I can't see any N/A mods creating enough extra flow to overpower stock injectors.
 
Without any extra air there is no reason to add extra fuel. If you are talking about doing a wild port job and serious cams, then the stock ECU is not friendly to that either, at that point a standalone is neccessary.

To answer your question no I do not believe the port fueler could be activated without positive pressure in the manifold, but I admit I have never used the interface so I do not know its full capabilities.
