How do I re-pressurize the fuel system so I can set my fuel pressure? The vacuum line is off, and both fuel lines are connected. I turned the key to the "ON" position without starting the car in hopes I'd hear the fuel pump come on and pressurize it. Nothing. Am I doing something wrong?

The fuel pump will not come on until you start cranking the car. You set the fuel pressure while the car is running.
 
Ok, that was my next course of action. I'm waiting on some parts to finish my head project, so I guess I'll just wait for everything to be put back together before I start it up and set the fuel pressure. Thanks!

True. Also, keep in mind that it will crank a bit before it actually starts.
