How hard or easy is it to tell, or not, if the cylinder is TDC. My manual that came with the tester talks about a whistle that tells you. Any ideas or pointers?

The way I do a CLT is hook it all up. Then turn the crank by hand you will know when the cylinder is at TDC you will not be able to turn the crank anymore it will start to push back but just hold it to get your reading.

I just look into the #1 spark plug and look to see where it is at and when it is going to fall from turning the crank either way a little bit.

So when it's TDC, Cylinder one and Three are ready. So would you recommend testing one and three first, crank it the other way and test two and four?
