My A/T takes a long time before shifting into the next gear and when I am in need to punch it, it'll make this loud knock when shifting to the next gear.

If I get a transmission flush, will this help it? Or would I need to just replace the current one?

First check and make sure it has enough fluid in it, and it doesn't smell burnt. Make sure you do so after you drive the car and it's all warmed up and run it through the gears and check it while in neutral. If that all looks good you might as get the fluid and filter changed before you go off and replace the tranny.

Most likely not. It's possible that you may just not have enough fluid in there, but I think there's a bigger internal problem, especially if it's been running with low fluid for a while.
